In this talk I will present current approaches for whole specimen tomography which take into account two different light propagation regimes: ballistic and diffusive. Depending on the amount of scattering present in the samples different approaches need to be taken in order to obtain 3D images of fluorophore distribution. During this talk emphasis will be given in the difference between direct and indirect imaging methods, showing practial examples of the difference between both in whole animal imaging experiments.
